I think it would be handy if there were a setting to allow e-mail alerts when someone quotes something you have said, using Reply and Quote, on the forums.

I think it is a good idea... but might be hard to track all the info needed to say "who posted to who... send message".

I say it would be very helpful, but can it be done?

Perhaps if the colour of the thread changed to show you someone is talking directly to you when you look at the thread list in the forum.

I'm sure it can be done. If it is an SQL database, and the Reply & Quote button calls an INSERT statement or stored procedure, then all the information is already present as you make the "quoted" post.
