How would it come to pass that he would become US president pro tempore?
VP Vance is currently the President of the Senate, so if he were to become POTUS, Chuck Grassley would become President of the Senate Pro-Tempore. It seems that latter position is decided within the Senate by vote, and currently Grassley holds that position.
If Trump, Vance, and Mike Johnson were somehow to be found unfit for office or unavailable to perform those duties, then Grassley would be next in line.
However, the ambitious Marco Rubio (Secretary of State) would be next in line after Grassley, and Secretary of the Treasury Scott Bessent would come after Rubio.

If Trump were to die or retire because of medical or legal issues, then JD Vance would be next in line. But if Trump and a bunch of his self-selected "loyal" members were removed from office on racketeering or organized-crime charges, then Grassley would be next. (I'll admit, such a scenario seems unlikely.)

Grassley's title is not US President of the Senate Pro-Tempore.
It is President Pro-Tempore of the Senate. It is similar to Speaker of the House, but is much more limited in scope. The Vice President runs the Senate (and is thus "President of the Senate" ), and the President Pro-Tem of the Senate takes over if the Vice President is unavailable, either through death or illness or temporary unavailability, such as if he's overseas. Although in that case, a temporary President Pro-Tem of the Senate is often chosen from among junior members of the party due to the position being mostly ceremonial, mainly presiding over the Senate, while the President Pro-Tem of the Senate usually has other duties (such as a committee chairman) and other significant demands on his time.
It seems rather complicated, but these rules are often covered in normal parliamentary procedure.

Sure, he's a Republican and a conservative, but I don't think he's MAGA.
Of the top people in the line of succession, I would trust Grassley much more than I would Vance, Johnson, or Rubio. I don't know about Bessent.
