best sites or software to upload video's to a web site?

It really depends on the amount of bandwidth you expect to use and the type of video files. For example, flash based video hosting is usually a lot more expensive than WMV/MOV.

If it's only a small scale operation, you're better off hosting the files on the same server you host the website. However, if you're running a business based on video streaming you're far better off getting a reliable company to handle all the video hosting for you.

Personally, I use these guys to host flash content for my video based sites. They're a bit pricey but incredibly reliable and have great customer service.
