Chris Rock, Steve Martin, Eddie Murphy, Rodney Dangerfield and Woody Allen would have to be my personal favourite stand-up comedians.

Comedy is very much an individual choice. I would go with the following:
Film - Life of Brian / Dr Strangelove (close, not sure which)
Comedian - Bill Bailey / Bill Hicks (for entirely different reasons)
Also worth mentioning Eddie Izzard, who is the funniest person ever not to insult someone, how he can be so funny without any form of malice is beyond me. And he does his routines in both French and English.
I would just say anything Monty Python, the Marx bros. definitely, laurel and Hardy, It's a mad, mad, mad, mad world and The Russians are coming.
