For my money, there are 3 best series out there:
Baldur's Gate Series
Neverwinter Nights Series
Morrowind Series
All have great character design elements, tonnes of side quests and a long, strong central plot. The best part about them is that all three have a unique interface and style.
Just in case anyone throws it out, I've played the Star Wars: KOTR, and while it was fun, it was WAYYY too easy and too short.
Edit: I've also tried Dungeon Siege... lots of fighting and good scenery... but there's no character development, a weak plot and OH MY GOD is it boring! Walk 3 feet, kill 10 things, walk 5 feet, kill 10 things... crap, is it ever tedious!
Monkey Island 1 and 2 just rock. Sierra adventure games seem somewhat frustrating and unimaginative compared to them, in my opinion. Haven't played any of the Monkey Island sequels after that, but I get the impression that they are inferior. I'd start with the first two.
I haven't played Sam and Max Hit the Road all the way through, but it's very funny. If you're interested in humorous adventure games that actually have humor in them, I'd recommend those three.
I've played about halfway through Grim Fandango, and while I don't think its dialogue is as sharp as in the other games I've mentioned, and some of its puzzles are irritating and counterintuitive, the graphics are lovely and the ambience can't be beat. That one's worth a look also.

I have to agree with the Myst-ers here!
Myst II (called RIVEN) is truly an incredible PC experience....awesome.
Myst III (EXILE) is also huge in scope...also awesome.
no guns, no dying....just pure logic, beauty, 3-D surround graphics to evoke nature. I feel like these Myst games are hearkening in a way that virtual reality gaming could be introduced and honed.

The first 2 Monkey Island games were classic beyond classic. The third one was good 'n funny, but the ending was lack-luster (to say the least). Never played the rest.
But I have to take issue with Myst. WTF? It's not an adventure game. Period. It's a puzzle game. The is no "role-playing", in the sense that you take on the "role" of an adventurer. There's just a putrid back story, some nice pictures, FMV and some "logic" puzzles. Feh.
