I'm not sure about films, but the best line ever was when Homer Simpson put on weight to get off work, and then ultimately saved the town from a nuclear explosion by getting jammed in the hole of the reactor.
Bart's comment, "I think it's ironic that for once dad's butt actually PREVENTED the escape of toxic gas!"
I didn't hear what was said after, for probably about two hours!

Steward (old Stewie): "That's never happened to me before."
Girl he slept with : "What? The five seconds of sex or the thirty minutes of crying."

Line by Clint Eastwood.
"Well you going to pull those pistols or you going to stand there and whistle Dixie"
Might have to be an American to understand the line.
Ex Confederate Soldier turned Outlaw when facing several Union Soldiers while outnumbered and upon being recognised. Dixie being a Confederate theme song hate ...[text shortened]... ght have done better to whistle Dixie.( But a little bit too big a slap in the face to ignore)

scary movie 3 "i can never do anything right, everyone hates me, i am never going to try and bring anyone back from the dead again"
good film
After I heard this, I was cracking up for days.
The Shoveler from Mystery Men: "We have a blind date with destiny and it looks like she just ordered the lobster."
From Unforgiven, after William Muny shoots a man:
"He had it comin'."
"We ALL have it comin', kid."
Sam Gamgee, in the rubble of the city of Osgiliath, in The Two Towers: "By rights, we shouldn't even be here, Mr. Frodo." In the books, Sam and Frodo never go to Osgiliath. Being a Tolkien snob, I laughed aloud.
